This assay has high sensitivity and excellent specificity for detection of GRA. No significant cross-reactivity or interference between GRA and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between GRA and all the analogues, therefore, cross reaction may still exist in some cases.

Store all reagents at 2-8 degree C.

We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. MBS726548 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Granulin (GRN) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GRN. The ELISA analytical biochemical technique of the MBS726548 kit is based on GRN antibody-GRN antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GRN antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GRN. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This GRA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse GRA. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: GRA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GRA antibody and an GRA-HRP conjugate. The assay sample and buffer are incubated together with GRA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GRA concentration since GRA from samples and GRA-HRP conjugate compete for the anti-GRA antibody . Since the number of sites is limited, as more sites are occupied by GRA from the sample, fewer sites are left to bind GRA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GRA concentration in each sample is interpolated from this standard curve.

Granulins are a family of secreted, glycosylated peptides that are cleaved from a single precursor protein with 7.5 repeats of a highly conserved 12-cysteine granulin/epithelin motif. The 88 kDa precursor protein, progranulin, is also called proepithelin and PC cell-derived growth factor. Cleavage of the signal peptide produces mature granulin which can be further cleaved into a variety of active, 6 kDa peptides. These smaller cleavage products are named granulin A, granulin B, granulin C, etc. Epithelins 1 and 2 are synonymous with granulins A and B, respectively. Both the peptides and intact granulin protein regulate cell growth. However, different members of the granulin protein family may act as inhibitors, stimulators, or have dual actions on cell growth. GRN: Granulins have possible cytokine-like activity. They may play a role in inflammation, wound repair, and tissue remodeling. Defects in GRN are the cause of ubiquitin-positive frontotemporal dementia (UP-FTD); also known as tau- negative frontotemporal dementia linked to chromosome 17. Frontotemporal dementia (FTD) is the second most common cause of dementia in people under the age of 65 years. It is an autosomal dominant neurodegenerative disease. Defects in GRN are the cause of neuronal ceroid lipofuscinosis type 11 (CLN11). A form of neuronal ceroid lipofuscinosis characterized by rapidly progressive visual loss due to retinal dystrophy, seizures, cerebellar ataxia, and cerebellar atrophy. Cognitive decline may also occur. Neuronal ceroid lipofuscinoses are progressive neurodegenerative, lysosomal storage diseases characterized by intracellular accumulation of autofluorescent liposomal material. Belongs to the granulin family. 2 isoforms of the human protein are produced by alternative splicing.
