MBS281166 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the 60S ribosomal protein L35a (RPL35A) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing RPL35A. The ELISA analytical biochemical technique of the MBS281166 kit is based on RPL35A antibody-RPL35A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect RPL35A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, RPL35A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate RPL35A in samples. An antibody specific for RPL35A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRPL35A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RPL35A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RPL35A bound in the initial step. The color development is stopped and the intensity of the color is measured.

RPL35A: Required for the proliferation and viability of hematopoietic cells. Plays a role in 60S ribosomal subunit formation. The protein was found to bind to both initiator and elongator tRNAs and consequently was assigned to the P site or P and A site. Defects in RPL35A are the cause of Diamond-Blackfan anemia type 5 (DBA5). DBA5 is a form of Diamond- Blackfan anemia, a congenital non-regenerative hypoplastic anemia that usually presents early in infancy. Diamond-Blackfan anemia is characterized by a moderate to severe macrocytic anemia, erythroblastopenia, and an increased risk of malignancy. 30 to 40% of Diamond-Blackfan anemia patients present with short stature and congenital anomalies, the most frequent being craniofacial (Pierre-Robin syndrome and cleft palate), thumb and urogenital anomalies. Belongs to the ribosomal protein L35Ae family.
