The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CDH16. The ELISA analytical biochemical technique of the MBS285139 kit is based on CDH16 antibody-CDH16 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CDH16 antigen targets in samples. Cadherin-16 is a member of the cadherin superfamily, genes encoding calcium-dependent, membrane-associated glycoproteins. Mapped to a previously identified cluster of cadherin genes on chromosome 16q22.1, the gene localizes with superfamily members CDH1, CDH3, CDH5, CDH8 and CDH11. The protein consists of an extracellular domain containing 6 cadherin domains, a transmembrane region and a truncated cytoplasmic domain but lacks the prosequence and tripeptide HAV adhesion recognition sequence typical of most classical cadherins. Expression is exclusively in kidney, where the protein functions as the principal mediator of homotypic cellular recognition, playing a role in the morphogenic direction of tissue development.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate CDH16 in samples. An antibody specific for CDH16 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCDH16 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CDH16 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CDH16 bound in the initial step. The color development is stopped and the intensity of the color is measured.
