This assay has high sensitivity and excellent specificity for detection of cTnI. No significant cross-reactivity or interference between cTnI and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between cTnI and all the analogues, therefore, cross reaction may still exist in some cases.

Store all reagents at 2-8 degree C.

We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. MBS754040 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Cardiac Troponin 1 (cTn1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing cTn1. The ELISA analytical biochemical technique of the MBS754040 kit is based on cTn1 antibody-cTn1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect cTn1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, cTn1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This cTnI ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine cTnI. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: cTnI ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-cTnI antibody and an cTnI-HRP conjugate. The assay sample and buffer are incubated together with cTnI-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the cTnI concentration since cTnI from samples and cTnI-HRP conjugate compete for the anti-cTnI antibody . Since the number of sites is limited, as more sites are occupied by cTnI from the sample, fewer sites are left to bind cTnI-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The cTnI concentration in each sample is interpolated from this standard curve.

Troponin I (TnI), along with troponin T (TnT) and troponin C (TnC), is one of 3 subunits that form the troponin complex of the thin filaments of striated muscle. TnI is the inhibitory subunit; blocking actin-myosin interactions and thereby mediating striated muscle relaxation. The TnI subfamily contains three genes: TnI-skeletal-fast-twitch, TnI-skeletal-slow-twitch, and TnI-cardiac. This gene encodes the TnI-cardiac protein and is exclusively expressed in cardiac muscle tissues.
