SDH5: a subunit of succinate dehydrogenase (SDH) complex II of the mitochondrial electron transport chain. SDH5 is required for insertion of FAD cofactor into SDHA, the catalytic subunit of succinate dehydrogenase (SDH) complex II. SDH is involved in complex II of the mitochondrial electron transport chain and is responsible for transferring electrons from succinate to ubiquinone (coenzyme Q). Mutated in some cancers. Germline mutations cause hereditary paragangliomas type 2, or non-chromaffin paragangliomas type 2. Paragangliomas are rare tumors that arise from components of the neuroendocrine system.

1. Loss of heterozygosity was found in more than 50 % of the von Hippel-Lindau-associated pheochromocytomas, and was correlated with a significant decrease (p < 0.05) in both SDHAF2 and SDHD mRNA expression, which may be suggestive of a pathogenic role.
