Principle of the Assay: This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with ACTH. During the reaction,ACTH in the sample or standard competes with a fixed amount ofACTH on the solid phase supporter for sites on the Biotinylated Detection
Antibody specific to ACTH. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin(SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of ACTH in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Processing yields several biologically active peptides, which are involved in diverse cellular functions, such as energy homeostasis, steroidogenesis, and increased melanin production in melanocytes. In mouse deficiency of this gene is associated with obesity, defects in adrenal development, and altered pigmentation. A pseudogene of this gene is located on chromosome 19.
