The Toll-like receptor (TLR) family of proteins are characterized by a highly conserved Toll homology (TH) domain; which is essential for Toll-induced signal transduction.TLR1; as well as the other TLR family members; are type I transmembrane receptors that characteristically contain an extracellular domain consisting of several leucine-rich regions along with a single cytoplasmic Toll/IL-1R-like domain. TLR2 and TLR4 are activated in response to lipopolysacchride (LPS) stimulation; which results in the activation and translocation of NFkB and suggests that these receptors are involved in mediating inflammatory responses. TLR6 is highly homologous to TLR1; and like other members of TLR family; it induces NFkB Signaling upon activation. Expression of TLR receptors is highest in peripheral blood leukocytes; macrophages; and monocytes.

This gene encodes a member of the toll-like receptor (TLR) family, which plays a fundamental role in pathogen recognition and activation of innate immune responses. These receptors recognize distinct pathogen-associated molecular patterns that are expressed on infectious agents. The protein encoded by this gene recognizes bacterial flagellin, the principal component of bacterial flagella and a virulence factor. The activation of this receptor mobilizes the nuclear factor NF-kappaB, which in turn activates a host of inflammatory-related target genes. Mutations in this gene have been associated with both resistance and susceptibility to systemic lupus erythematosus, and susceptibility to Legionnaire disease.[provided by RefSeq, Dec 2009]
