The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing AS3MT. The ELISA analytical biochemical technique of the MBS2706727 kit is based on AS3MT antibody-AS3MT antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AS3MT antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AS3MT. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Arsenic, 3-Oxidation State Methyltransferase (AS3MT). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Arsenic, 3-Oxidation State Methyltransferase (AS3MT). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Arsenic, 3-Oxidation State Methyltransferase (AS3MT), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Arsenic, 3-Oxidation State Methyltransferase (AS3MT) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

AS3MT catalyzes the transfer of a methyl group from S-adenosyl-L-methionine (AdoMet) to trivalent arsenical and may play a role in arsenic metabolism (Lin et al., 2002 [PubMed 11790780]).[supplied by OMIM, Mar 2008]

Catalyzes the transfer of a methyl group from AdoMet to trivalent arsenicals producing methylated and dimethylated arsenicals. It methylates arsenite to form methylarsonate, Me-AsO3H2, which is reduced by methylarsonate reductase to methylarsonite, Me-As(OH)2. Methylarsonite is also a substrate and it is converted into the much less toxic compound dimethylarsinate (cacodylate), Me2As(O)-OH ().
