This assay has high sensitivity and excellent specificity for detection of SOX3. No significant cross-reactivity or interference between SOX3 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SOX3 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses
This SOX3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep SOX3. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. Principle of the assay: SOX3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SOX3 antibody and an SOX3-HRP conjugate. The assay sample and buffer are incubated together with SOX3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SOX3 concentration since SOX3 from samples and SOX3-HRP conjugate compete for the anti-SOX3 antibody . Since the number of sites is limited, as more sites are occupied by SOX3 from the sample, fewer sites are left to bind SOX3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SOX3 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. The encoded protein may act as a transcriptional regulator after forming a protein complex with other proteins. SOX3: Transcription factor required during the formation of the hypothalamo-pituitary axis. May function as a switch in neuronal development. Keeps neural cells undifferentiated by counteracting the activity of proneural proteins and suppresses neuronal differentiation. Required also within the pharyngeal epithelia for craniofacial morphogenesis. Controls a genetic switch in male development. Is necessary for initiating male sex determination by directing the development of supporting cell precursors (pre-Sertoli cells) as Sertoli rather than granulosa cells. Defects in SOX3 are a cause of panhypopituitarism X- linked (PHPX). Affected individuals have absent infundibulum, anterior pituitary hypoplasia, and ectopic posterior pituitary. Defects in SOX3 are the cause of mental retardation X- linked with isolated growth hormone deficiency (MRXGH). Defects in SOX3 are the cause of 46,XX sex reversal type 3 (SRXX3). A condition in which male gonads develop in a genetic female (female to male sex reversal). Copy number variations (CNV) encompassing or in close proximity to SOX3 are responsible for XX male reversal. These variations include two duplications of approximately 123 kb and 85 kb, the former of which spans the entire SOX3 gene; a 343 kb deletion immediately upstream of SOX3 that is probably responsible of altered regulation (and not increased dosage) of SOX3; a large (approximately 6 Mb) duplication that encompasses SOX3 and at least 18 additional distally located genes. Its proximal breakpoint falls within the SOX3 regulatory region. This large rearrangement has been found in a patient with XX male reversal and a complex phenotype that also includes a scrotal hypoplasia, microcephaly, developmental delay, and growth retardation.
