Specificity
This assay has high sensitivity and excellent specificity for detection of SCNN1G. No significant cross-reactivity or interference between SCNN1G and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SCNN1G and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Select online data sheet information is drawn from bioinformatics databases, occasionally resulting in ambiguous or non-relevant product information. It is the responsibility of the customer to review, verify, and evaluate the information to make sure it matches their requirements before purchasing the kit. Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of SCNN1G elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS7217895 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Amiloride sensitive sodium channel subunit gamma (SCNN1G) ELISA Kit target analytes in
biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing SCNN1G. The ELISA analytical biochemical technique of the MBS7217895 kit is based on SCNN1G antibody-SCNN1G antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SCNN1G antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SCNN1G. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This SCNN1G ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human SCNN1G. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
Principle of the Assay: SCNN1G ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-SCNN1G antibody and an SCNN1G-HRP conjugate. The assay sample and buffer are incubated together with SCNN1G-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SCNN1G concentration since SCNN1G from samples and SCNN1G-HRP conjugate compete for the anti-SCNN1G antibody . Since the number of sites is limited, as more sites are occupied by SCNN1G from the sample, fewer sites are left to bind SCNN1G-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SCNN1G concentration in each sample is interpolated from this standard curve.

NCBI Summary for SCNN1G
Nonvoltage-gated, amiloride-sensitive, sodium channels control fluid and electrolyte transport across epithelia in many organs. These channels are heteromeric complexes consisting of 3 subunits: alpha, beta, and gamma. This gene encodes the gamma subunit, and mutations in this gene have been associated with Liddle syndrome. UniProt Comments for SCNN1G
ENaC-gamma: Sodium permeable non-voltage-sensitive ion channel inhibited by the diuretic amiloride. Mediates the electrodiffusion of the luminal sodium (and water, which follows osmotically) through the apical membrane of epithelial cells. Controls the reabsorption of sodium in kidney, colon, lung and sweat glands. Also plays a role in taste perception. Defects in SCNN1G are a cause of Liddle syndrome (LIDDS). It is an autosomal dominant disorder characterized by pseudoaldosteronism and hypertension associated with hypokalemic alkalosis. The disease is caused by constitutive activation of the renal epithelial sodium channel. Defects in SCNN1G are the cause of bronchiectasis with or without elevated sweat chloride type 3 (BESC3). A debilitating respiratory disease characterized by chronic, abnormal dilatation of the bronchi and other cystic fibrosis-like symptoms in the absence of known causes of bronchiectasis (cystic fibrosis, autoimmune diseases, ciliary dyskinesia, common variable immunodeficiency, foreign body obstruction). Clinical features include sub-normal lung function, sinopulmonary infections, chronic productive cough, excessive sputum production, and elevated sweat chloride in some cases. Belongs to the amiloride-sensitive sodium channel (TC 1.A.6) family. SCNN1G subfamily.
