This assay has high sensitivity and excellent specificity for detection of CASR. No significant cross-reactivity or interference between CASR and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CASR and all the analogues, therefore, cross reaction may still exist in some cases.

Store all reagents at 2-8 degree C.

We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. MBS729896 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Calcium Sensing Receptor (CaSR) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CaSR. The ELISA analytical biochemical technique of the MBS729896 kit is based on CaSR antibody-CaSR antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CaSR antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CaSR. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: CASR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CASR antibody and an CASR-HRP conjugate. The assay sample and buffer are incubated together with CASR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CASR concentration since CASR from samples and CASR-HRP conjugate compete for the anti-CASR antibody . Since the number of sites is limited, as more sites are occupied by CASR from the sample, fewer sites are left to bind CASR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CASR concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a G protein-coupled receptor that is expressed in the parathyroid hormone (PTH)-producing chief cells of the parathyroid gland, and the cells lining the kidney tubule. It senses small changes in circulating calcium concentration and couples this information to intracellular signaling pathways that modify PTH secretion or renal cation handling, thus this protein plays an essential role in maintaining mineral ion homeostasis. Mutations in this gene cause familial hypocalciuric hypercalcemia, familial, isolated hypoparathyroidism, and neonatal severe primary hyperparathyroidism. [provided by RefSeq, Jul 2008]
