The divalent cations (Ca2+ and Mg2+) are important for a multitude of cellular functions. A G-protein coupled receptor, termed calcium sensing receptor, responds to extracellular calcium and mediates several of the known effects of Ca on parathyroid and renal functions. CaSR is expressed in a number of tissues including thyroid C-cells,. Brain, and GI tract. Ca2+ acts as first messenger via CaSR-mediated activation of second messenger systems (e.g., phospholipase C and A2, cAMP) altering cellular physiology. The protein encoded by this gene is a G protein-coupled receptor that is expressed in the parathyroid hormone (PTH)-producing chief cells of the parathyroid gland, and the cells lining the kidney tubule. It senses small changes in circulating calcium concentration and couples this information to intracellular signaling pathways that modify PTH secretion or renal cation handling, thus this protein plays an essential role in maintaining mineral ion homeostasis. Mutations in this gene cause familial hypocalciuric hypercalcemia, familial, isolated hypoparathyroidism, and neonatal severe primary hyperparathyroidism. [provided by RefSeq]
