Intended Uses: This immunoassay kit allows for the in vitro quantitative determination of human 1-phosphatidylinositol-4,5-bisphosphate phosphodiesterase gamma-1 concentrations in serum, Plasma, tissue homogenates and Cell culture supernates and Other biological fluids.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to 1-phosphatidylinositol-4,5-bisphosphate phosphodiesterase gamma-1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for 1-phosphatidylinositol-4,5-bisphosphate phosphodiesterase gamma-1 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain 1-phosphatidylinositol-4,5-bisphosphate phosphodiesterase gamma-1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of 1-phosphatidylinositol-4,5-bisphosphate phosphodiesterase gamma-1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene catalyzes the formation of inositol 1,4,5-trisphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate. This reaction uses calcium as a cofactor and plays an important role in the intracellular transduction of receptor-mediated tyrosine kinase activators. For example, when activated by SRC, the encoded protein causes the Ras guanine nucleotide exchange factor RasGRP1 to translocate to the Golgi, where it activates Ras. Also, this protein has been shown to be a major substrate for heparin-binding growth factor 1 (acidic fibroblast growth factor)-activated tyrosine kinase. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
