biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TRPV4. The ELISA analytical biochemical technique of the MBS700415 kit is based on TRPV4 antibody-TRPV4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TRPV4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TRPV4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique.
Antibody specific for TrpV4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TrpV4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TrpV4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TrpV4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the OSM9-like transient receptor potential channel (OTRPC) subfamily in the transient receptor potential (TRP) superfamily of ion channels. The encoded protein is a Ca2+-permeable, nonselective cation channel that is thought to be involved in the regulation of systemic osmotic pressure. Mutations in this gene are the cause of spondylometaphyseal and metatropic dysplasia and hereditary motor and sensory neuropathy type IIC. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Apr 2010]

Activation by exposure to hypotonicity within the physiological range exhibits an outward rectification. Also activated by low pH, citrate and phorbol esters. Increase of intracellular Ca2+ potentiates currents. Channel activity seems to be regulated by a calmodulin-dependent mechanism with a negative feedback mechanism. Promotes cell-cell junction formation in skin keratinocytes and plays an important role in the formation and/or maintenance of functional intercellular barriers. Acts as a regulator of intracellular Ca2+ in synoviocytes.
