MBS751061 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Anti-Type II Collagen Antibody (CII) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CII. The ELISA analytical biochemical technique of the MBS751061 kit is based on CII antibody-CII antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CII antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CII. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This Anti-CII ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Goat Anti-CII. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: Anti-CII ELISA kit applies the competitive enzyme immunoassay technique utilizing Type II Collagen antigen and an Anti-CII-HRP conjugate. The assay sample and buffer are incubated together with Anti-CII-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Anti-CII concentration since Anti-CII from samples and Anti-CII-HRP conjugate compete for the Type II Collagen antigen . Since the number of sites is limited, as more sites are occupied by Anti-CII from the sample, fewer sites are left to bind Anti-CII-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Anti-CII concentration in each sample is interpolated from this standard curve.

Mutations in this gene are associated with achondrogenesis, chondrodysplasia, early onset familial osteoarthritis, SED congenita, Langer-Saldino achondrogenesis, Kniest dysplasia, Stickler syndrome type I, and spondyloepimetaphyseal dysplasia Strudwick type. In addition, defects in processing chondrocalcin, a calcium binding protein that is the C-propeptide of this collagen molecule, are also associated with chondrodysplasia.
