Specificity
This assay has high sensitivity and excellent specificity for detection of FBLN. No significant cross-reactivity or interference between FBLN and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FBLN and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Select online data sheet information is drawn from bioinformatics databases, occasionally resulting in ambiguous or non-relevant product information. It is the responsibility of the customer to review, verify, and evaluate the information to make sure it matches their requirements before purchasing the kit. Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of FBLN elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS751885 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Fibulin (FBLN) ELISA Kit target analytes in
biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing FBLN. The ELISA analytical biochemical technique of the MBS751885 kit is based on FBLN antibody-FBLN antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect FBLN antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, FBLN. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: FBLN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FBLN antibody and an FBLN-HRP conjugate. The assay sample and buffer are incubated together with FBLN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FBLN concentration since FBLN from samples and FBLN-HRP conjugate compete for the anti-FBLN antibody . Since the number of sites is limited, as more sites are occupied by FBLN from the sample, fewer sites are left to bind FBLN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FBLN concentration in each sample is interpolated from this standard curve.

NCBI Summary for FBLN
Fibulin 1 is a secreted glycoprotein that becomes incorporated into a fibrillar extracellular matrix. Calcium-binding is apparently required to mediate its binding to laminin and nidogen. It mediates platelet adhesion via binding fibrinogen. Four splice variants which differ in the 3' end have been identified. Each variant encodes a different isoform, but no functional distinctions have been identified among the four variants. [provided by RefSeq, Jul 2008]
UniProt Comments for FBLN
FBLN1: Incorporated into fibronectin-containing matrix fibers. May play a role in cell adhesion and migration along protein fibers within the extracellular matrix (ECM). Could be important for certain developmental processes and contribute to the supramolecular organization of ECM architecture, in particular to those of basement membranes. Has been implicated in a role in cellular transformation and tumor invasion, it appears to be a tumor suppressor. May play a role in haemostasis and thrombosis owing to its ability to bind fibrinogen and incorporate into clots. Could play a significant role in modulating the neurotrophic activities of APP, particularly soluble APP. A chromosomal aberration involving FBLN1 is found in a complex type of synpolydactyly referred to as 3/3-prime/4 synpolydactyly associated with metacarpal and metatarsal synostoses. Reciprocal translocation t(12;22)(p11.2;q13.3) with C12orf2. Fibroblasts derived from a patient with synpolydactyly displayed alterations in the level of isoform D splice variant incorporated into the ECM and secreted into the conditioned culture medium. By contrast, the expression of isoform C was not perturbed in the patients fibroblasts. Furthermore, no aberrant polypeptides were detected in extracts of cultured patients fibroblasts. The translocation t(12;22) may result in haploinsufficiency of the isoform D splice variant, which could lead to the observed limb malformation. Elevated expression and altered processing of FBLN1 protein is associated with human breast cancer. Belongs to the fibulin family. 4 isoforms of the human protein are produced by alternative splicing.

1. FBLN1 functions as a novel candidate tumor suppressor gene in CM, and its downregulation may be due to promoter hypermethylation.
