Specificity
This assay has high sensitivity and excellent specificity for detection of BTA. No significant cross-reactivity or interference between BTA and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between BTA and all the analogues, therefore, cross reaction may still exist in some cases.

Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team

Principle of the assay: BTA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BTA antibody and an BTA-HRP conjugate. The assay sample and buffer are incubated together with BTA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BTA concentration since BTA from samples and BTA-HRP conjugate compete for the anti-BTA antibody . Since the number of sites is limited, as more sites are occupied by BTA from the sample, fewer sites are left to bind BTA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BTA concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the heparan sulfate/heparin GlcNAc N-deacetylase/ N-sulfotransferase family. The encoded enzyme is a type II transmembrane protein that resides in the Golgi apparatus. The encoded protein catalyzes the transfer of sulfate from 3'-phosphoadenosine 5'-phosphosulfate to nitrogen of glucosamine in heparan sulfate. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2014]
