Flow Cytometry
1. T98G cells. may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. After blocking, add 10ul of D2727-70C to <=1x10e6 cells and incubated for 30 minutes at RT.
3. Wash the cells twice in MBS635370 Flow Cytometry Staining Buffer. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer.
4. Resuspended the cells in MBS635370 Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled mouse IgG2B antibody. This procedure may need to be modified, depending upon the cell type and final utilization. Individual users may need to titrate to determine the optimal reagent amount for their specific use.
Preparation and Storage
May be stored at 4 degree C before opening. DO NOT FREEZE! Stable at 4 degree C. Stable for 12 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Freezing APC conjugates will result in a substantial loss of activity. APC conjugates are sensitive to light.
Other Notes
Small volumes of anti-DLL3 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Related Product Information for
anti-DLL3 antibody
DLL1 is the human homolog of the Drosophila Notch ligand, Delta. It is a type I transmembrane protein with a DSL (Delta, Serrate, Jagged) domain followed by 8 EGF-like domains in its extracellular region and a short cytoplasmic region. Notch-DSL signaling plays an important role in the differentiation processes of many tissues.

NCBI Summary for DLL3
This gene encodes a member of the delta protein ligand family. This family functions as Notch ligands that are characterized by a DSL domain, EGF repeats, and a transmembrane domain. Mutations in this gene cause autosomal recessive spondylocostal dysostosis 1. Two transcript variants encoding distinct isoforms have been identified for this gene. [provided by RefSeq, Jul 2008]
UniProt Comments for DLL3
DLL3: Inhibits primary neurogenesis. May be required to divert neurons along a specific differentiation pathway. Plays a role in the formation of somite boundaries during segmentation of the paraxial mesoderm. Can bind and activate Notch-1 or another Notch receptor.
Protein type: Membrane protein, integral; Cell development/differentiation
Chromosomal Location of Human Ortholog: 19q13
Cellular Component: integral to membrane
Molecular Function: Notch binding
Biological Process: paraxial mesoderm development; compartment specification; somitogenesis; Notch signaling pathway; skeletal development; negative regulation of neurogenesis
Disease: Spondylocostal Dysostosis 1, Autosomal Recessive
Research Articles on DLL3
1. DLL3 was silenced by methylation in human human hepatocellular carcinoma and it negatively regulates the growth of human hepatocellular carcinoma cells.
