The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CRLF2. The ELISA analytical biochemical technique of the MBS166445 kit is based on CRLF2 antibody-CRLF2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CRLF2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CRLF2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This sandwich kit is for the accurate quantitative detection of Human Cytokine receptor-like factor 2 (also known as CRLF2) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human CRLF2 antibody. CRLF2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CRLF2 Antibody is added and binds to CRLF2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CRLF2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CRLF2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

This gene encodes a member of the type I cytokine receptor family. The encoded protein is a receptor for thymic stromal lymphopoietin (TSLP). Together with the interleukin 7 receptor (IL7R), the encoded protein and TSLP activate STAT3, STAT5, and JAK2 pathways, which control processes such as cell proliferation and development of the hematopoietic system. Rearrangement of this gene with immunoglobulin heavy chain gene (IGH) on chromosome 14, or with P2Y purinoceptor 8 gene (P2RY8) on the same X or Y chromosomes is associated with B-progenitor acute lymphoblastic leukemia (ALL) and Down syndrome ALL.
