The ELISA analytical biochemical technique of the MBS2881897 kit is based on Bcl2 antibody-Bcl2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Bcl2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Bcl2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Intended Uses: This immunoassay kit allows for the in vitro quantitative determination of mouse Apoptosis regulator Bcl-2 concentrations in serum, Plasma, tissue homogenates and Cell culture supernates and Other biological fluids.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Apoptosis regulator Bcl-2. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Apoptosis regulator Bcl-2 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Apoptosis regulator Bcl-2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Apoptosis regulator Bcl-2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the B cell lymphoma 2 protein family. Members of this family regulate cell death in multiple cell types and can have either proapoptotic or antiapoptotic activities. The protein encoded by this gene inhibits mitochondrial-mediated apoptosis. This protein is an integral outer mitochondrial membrane protein that functions as part of signaling pathway that controls mitochondrial permeability in response to apoptotic stimuli. This protein may also play a role in neuron cell survival and autophagy. Abnormal expression and chromosomal translocations of this gene are associated with cancer progression in numerous tissues. 1. The expression of GRP78 and Bcl-2 was significantly higher in astrocytes pretreated with recombinant Shh. These findings suggest that the expression of Shh may inhibit cell death by activating Bcl-2 through a GRP78-dependent pathway.
