MBS720605 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Acid Phosphatase 5, TartRat e Resistant (ACP5) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ACP5. The ELISA analytical biochemical technique of the MBS720605 kit is based on ACP5 antibody-ACP5 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ACP5 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ACP5. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: TRACP5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TRACP5 antibody and an TRACP5-HRP conjugate. The assay sample and buffer are incubated together with TRACP5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TRACP5 concentration since TRACP5 from samples and TRACP5-HRP conjugate compete for the anti-TRACP5 antibody . Since the number of sites is limited, as more sites are occupied by TRACP5 from the sample, fewer sites are left to bind TRACP5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TRACP5 concentration in each sample is interpolated from this standard curve.

Its expression seems to increase in certain pathological states such as Gaucher and Hodgkin diseases, the hairy cell, the B-cell, and the T-cell leukemias. Defects in ACP5 are the cause of spondyloenchondrodysplasia with immune dysregulation (SPENCDI). A disease characterized by vertebral and metaphyseal dysplasia, spasticity with cerebral calcifications, and strong predisposition to autoimmune diseases. The skeletal dysplasia is characterized by radiolucent and irregular spondylar and metaphyseal lesions that represent islands of chondroid tissue within bone. ACP5 inactivating mutations result in a functional excess of phosphorylated osteopontin causing deregulation of osteopontin signaling and consequential autoimmune disease. Belongs to the metallophosphoesterase superfamily. Purple acid phosphatase family.
