MBS564053 is a ready-to-use microwell, strip-or-full plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Human Lactoferrin (LTF) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LTF. The ELISA analytical biochemical technique of the MBS564053 kit is based on LTF antibody-LTF antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LTF antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LTF. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%). Intended Uses: The Lactoferrin test kit is a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring Lactoferrin in human biological samples.

Principle of the Assay: The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the LF present in samples reacts with the anti-LF antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-LF antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound LF. Following another washing step, the enzyme bound to the Immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of LF in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of LF in the test sample. The quantity of LF in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Background: Lactoferrin (LF) is a multifunctional iron-binding protein with antimicrobial activities. It is found in milk and other mucosal secretions. Lactoferrin is known to be part of the innate defense against infection and inflammation and also has a function in regulation of iron absorption.
