This assay has high sensitivity and excellent specificity for detection of AK1. No significant cross-reactivity or interference between AK1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between AK1 and all the analogues, therefore, cross reaction may still exist in some cases.

Store all reagents at 2-8 degree C.

We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing AK1. The ELISA analytical biochemical technique of the MBS7201272 kit is based on AK1 antibody-AK1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AK1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AK1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This AK1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human AK1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: AK1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AK1 antibody and an AK1-HRP conjugate. The assay sample and buffer are incubated together with AK1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AK1 concentration since AK1 from samples and AK1-HRP conjugate compete for the anti-AK1 antibody . Since the number of sites is limited, as more sites are occupied by AK1 from the sample, fewer sites are left to bind AK1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AK1 concentration in each sample is interpolated from this standard curve.

Adenylate kinase is an enzyme involved in regulating the adenine nucleotide composition within a cell by catalyzing the reversible transfer of phosphate group among adinine nucleotides. Three isozymes of adenylate kinase have been identified in vertebrates, adenylate isozyme 1 (AK1), 2 (AK2) and 3 (AK3). AK1 is found in the cytosol of skeletal muscle, brain and erythrocytes, whereas AK2 and AK3 are found in the mitochondria of other tissues including liver and heart. AK1 was identified because of its association with a rare genetic disorder causing nonspherocytic hemolytic anemia where a mutation in the AK1 gene was found to reduce the catalytic activity of the enzyme.
