The ELISA analytical biochemical technique of the MBS764643 kit is based on NPHN antibody-NPHN antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect NPHN antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, NPHN. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. anti-Rat NPHN antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Rat NPHN antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Rat NPHN amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration ofRat NPHN can be calculated.

This gene encodes a member of the immunoglobulin family of cell adhesion molecules that functions in the glomerular filtration barrier in the kidney. The gene is primarily expressed in renal tissues, and the protein is a type-1 transmembrane protein found at the slit diaphragm of glomerular podocytes. The slit diaphragm is thought to function as an ultrafilter to exclude albumin and other plasma macromolecules in the formation of urine. Mutations in this gene result in Finnish-type congenital nephrosis 1, characterized by severe proteinuria and loss of the slit diaphragm and foot processes.[provided by RefSeq, Oct 2009]
