MBS1751113 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the BMP-7, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing BMP-7. The ELISA analytical biochemical technique of the MBS1751113 kit is based on BMP-7 antibody-BMP-7 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect BMP-7 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, BMP-7. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: 215 Chinese Hamster BMP-7 ELISA Kit was based on standard sandwich enzyme-linked immunesorbent assay technology. A specific for Chinese Hamster BMP-7 has been precoated onto 96-well plates. Standards(Expression system for standard: CHO; Immunogen sequence: S292-H430) and test samples are added to the wells, a biotinylated detection specific for Chinese Hamster BMP-7 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the 215 Chinese Hamster BMP-7 amount of sample captured in plate.

Background: Bone morphogenetic protein 7 or BMP7 (also known as osteogenic protein-1 or OP-1) is a protein that in humans is encoded by the BMP7 gene.The protein encoded by this gene is a member of the TGF-beta superfamily. Like other members of the bone morphogenetic protein family of proteins, it plays a key role in the transformation of mesenchymal cells into bone and cartilage. It is inhibited by noggin and a similar protein, chordin, which are expressed in the Spemann-Mangold Organizer. BMP7 may be involved in bone homeostasis. It is expressed in the brain, kidneys and bladder.

This gene encodes a secreted ligand of the TGF-beta (transforming growth factor-beta) superfamily of proteins. Ligands of this family bind various TGF-beta receptors leading to recruitment and activation of SMAD family transcription factors that regulate gene expression. The encoded preproprotein is proteolytically processed to generate each subunit of the disulfide-linked homodimer. Mutation of this gene results in skeletal, kidney, and other developmental defects. Induces cartilage and bone formation. May be the osteoinductive factor responsible for the phenomenon of epithelial osteogenesis. Plays a role in calcium regulation and bone homeostasis.
