The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing NPS. The ELISA analytical biochemical technique of the MBS2700290 kit is based on NPS antibody-NPS antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect NPS antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, NPS. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to Neuropeptide S (NPS) has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled Neuropeptide S (NPS) and unlabeled Neuropeptide S (NPS) (Standards or samples) with the pre-coated antibody specific to Neuropeptide S (NPS). After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of Neuropeptide S (NPS) in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of Neuropeptide S (NPS) in the sample.
