Intended Uses: This CYP2C8 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CYP2C8. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: CYP2C8 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CYP2C8 antibody and an CYP2C8-HRP conjugate. The assay sample and buffer are incubated together with CYP2C8-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CYP2C8 concentration since CYP2C8 from samples and CYP2C8-HRP conjugate compete for the anti-CYP2C8 antibody . Since the number of sites is limited, as more sites are occupied by CYP2C8 from the sample, fewer sites are left to bind CYP2C8-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CYP2C8 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and its expression is induced by phenobarbital. The enzyme is known to metabolize many xenobiotics, including the anticonvulsive drug mephenytoin, benzo(a)pyrene, 7-ethyoxycoumarin, and the anti-cancer drug taxol. This gene is located within a cluster of cytochrome P450 genes on chromosome 10q24. Several transcript variants encoding a few different isoforms have been found for this gene. CYP2C8: Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It oxidizes a variety of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. In the epoxidation of arachidonic acid it generates only 14,15- and 11,12-cis-epoxyeicosatrienoic acids. It is the principal enzyme responsible for the metabolism the anti- cancer drug paclitaxel (taxol).
