Specificity
This assay has high sensitivity and excellent specificity for detection of human NCAM-L1. No significant cross-reactivity or interference between human NCAM-L1 and analogues was observed.

Sensitivity
The minimum detectable dose of human NCAM-L1 is typically less than 0.31 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision
Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing L1CAM. The ELISA analytical biochemical technique of the MBS700933 kit is based on L1CAM antibody-L1CAM antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect L1CAM antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, L1CAM. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Princple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique.
Antibody specific for NCAM-L1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NCAM-L1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NCAM-L1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NCAM-L1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is an axonal glycoprotein belonging to the immunoglobulin supergene family. The ectodomain, consisting of several immunoglobulin-like domains and fibronectin-like repeats (type III), is linked via a single transmembrane sequence to a conserved cytoplasmic domain. This cell adhesion molecule plays an important role in nervous system development, including neuronal migration and differentiation. Mutations in the gene cause X-linked neurological syndromes known as CRASH (corpus callosum hypoplasia, retardation, aphasia, spastic paraplegia and hydrocephalus). Alternative splicing of this gene results in multiple transcript variants, some of which include an alternate exon that is considered to be specific to neurons. [provided by RefSeq, May 2013]
