The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing NKX3-1. The ELISA analytical biochemical technique of the MBS2706217 kit is based on NKX3-1 antibody-NKX3-1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect NKX3-1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, NKX3-1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to NK3 Homeobox Protein 1 (NKX3-1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to NK3 Homeobox Protein 1 (NKX3-1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain NK3 Homeobox Protein 1 (NKX3-1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of NK3 Homeobox Protein 1 (NKX3-1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a homeobox-containing transcription factor. This transcription factor functions as a negative regulator of epithelial cell growth in prostate tissue. Aberrant expression of this gene is associated with prostate tumor progression. Alternate splicing results in multiple transcript variants of this gene. [provided by RefSeq, Jan 2012]

Transcription factor, which binds preferentially the consensus sequence 5'-TAAGT[AG]-3' and can behave as a transcriptional repressor. Plays an important role in normal prostate development, regulating proliferation of glandular epithelium and in the formation of ducts in prostate. Acts as a tumor suppressor controlling prostate carcinogenesis, as shown by the ability to inhibit proliferation and invasion activities of PC-3 prostate cancer cells.
