MBS1751131 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the BMP-6, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing BMP-6. The ELISA analytical biochemical technique of the MBS1751131 kit is based on BMP-6 antibody-BMP-6 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect BMP-6 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, BMP-6. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: Rat BMP-6 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A a monoclonal antibody from mouse specific for BMP-6 has been precoated onto 96- well plates. Standards(Expression system for standard: CHO, Immunogen sequence: S368-H506) and test samples are added to the wells, a biotinylated detection a biotinylated polyclonal antibody from goat specific for BMP-6 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Rat BMP-6 amount of sample captured in plate.

Background: The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, BMP-6 has a proposed role in early development. In addition, the fact that this BMP is closely related to BMP5 and BMP7 has lead to speculation of possible bone inductive activity.
