MBS920884 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the apolipoprotein L, 1 (APOL1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing APOL1. The ELISA analytical biochemical technique of the MBS920884 kit is based on APOL1 antibody-APOL1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect APOL1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, APOL1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with APOL1. Standards or samples are then added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) -conjugated antibody preparation specific for APOL1C mix well and incubated. The more the amount of APOL1 in samples, the less HRP-antibody bound by pre-coated APOL1. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. And the color develops in opposite to the amount of APOL1 in the sample. The color development is stopped and the intensity of the color is measured.

This gene encodes a secreted high density lipoprotein which binds to apolipoprotein A-I. Apolipoprotein A-I is a relatively abundant plasma protein and is the major apoprotein of HDL. It is involved in the formation of most cholesteryl esters in plasma and also promotes efflux of cholesterol from cells. This apolipoprotein L family member may play a role in lipid exchange and transport throughout the body, as well as in reverse cholesterol transport from peripheral cells to the liver. APOL1: May play a role in lipid exchange and transport throughout the body. May participate in reverse cholesterol transport from peripheral cells to the liver. Defects in APOL1 are the cause of focal segmental glomerulosclerosis type 4 (FSGS4). It is a renal pathology defined by the presence of segmental sclerosis in glomeruli and resulting in proteinuria, reduced glomerular filtration rate and edema. Renal insufficiency often progresses to end-stage renal disease, a highly morbid state requiring either dialysis therapy or kidney transplantation. Belongs to the apolipoprotein L family. 2 isoforms of the human protein are produced by alternative splicing.
