MBS262050 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the thymidylate synthase (TS) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TS. The ELISA analytical biochemical technique of the MBS262050 kit is based on TS antibody-TS antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TS antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TS. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Thymidylate synthase catalyzes the methylation of deoxyuridylate to deoxythymidylate using 5,10-methylenetetrahydrofolate (methylene-THF) as a cofactor. This function maintains the dTMP (thymidine-5-prime monophosphate) pool critical for DNA replication and repair. The enzyme has been of interest as a target for cancer chemotherapeutic agents. It is considered to be the primary site of action for 5-fluorouracil, 5-fluoro-2-prime-deoxyuridine, and some folate analogs. Expression of this gene and that of a naturally occuring antisense transcript rTSalpha (GeneID:55556) vary inversely when cell-growth progresses from late-log to plateau phase. TYMS: an enzyme in the thymidylate synthase family. Polymorphisms serve as prognostic markers for determining modality of treatment in certain cancers. Genotyping helps predict toxicity to 5-FU-based chemotherapy in patients with colorectal cancer and the response to platinum-based chemotherapy in advanced NSCLC.
