The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TYMS. The ELISA analytical biochemical technique of the MBS450515 kit is based on TYMS antibody-TYMS antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TYMS antigen targets in samples. Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of TYMS in human serum, plasma, tissue homogenates and other biological fluids.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to TYMS. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to TYMS. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain TYMS, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of TYMS in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Thymidylate synthase catalyzes the methylation of deoxyuridylate to deoxythymidylate using 5,10-methylenetetrahydrofolate (methylene-THF) as a cofactor. This function maintains the dTMP (thymidine-5-prime monophosphate) pool critical for DNA replication and repair. The enzyme has been of interest as a target for cancer chemotherapeutic agents. It is considered to be the primary site of action for 5-fluorouracil, 5-fluoro-2-prime-deoxyuridine, and some folate analogs. Expression of this gene and that of a naturally occuring antisense transcript rTSalpha (GeneID:55556) vary inversely when cell-growth progresses from late-log to plateau phase.
