Applications Tested/Suitable for anti-TSC1 antibody
Peptide ELISA (EIA), Western Blot (WB), Immunohistochemistry (IHC)
Application Notes for anti-TSC1 antibody
Peptide ELISA: Antibody detection limit dilution 1: 128000.
Immunohistochemistry: In paraffin embedded Human Uterus shows membranous staining around non-basal nuclei and at the lumen of the endometrial gland. Recommended concentration, 3-6ug/ml.
Western Blot: Approx 150kDa band observed in Human Brain lysates (calculated MW of 130kDa according to NP_000359.1). The observed molecular weight corresponds to earlier findings in literature with different antibodies (Miloloza et al Hum Mol Genet.2000 Jul 22;9(12): 1721-7; PMID: 10915759). In transfected HEK293 transiently expressing Human TSC1 a band of approx.120kDa is observed. This band is not observed in the non-transfected HEK293. Recommended concentration: 0.1-0.3ug/ml.
Western Blot (WB) of anti-TSC1 antibody
(0.1ug/ml) staining of Human Brain lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.

Western Blot (WB) of anti-TSC1 antibody
HEK293 overexpressing Human TSC1 (RC213332) and probed (mock transfection in first lane), tested by Origene.

Immunohistochemistry (IHC) of anti-TSC1 antibody
(3.8ug/ml) staining of paraffin embedded Human Uterus. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.

NCBI Summary for TSC1
This gene encodes a growth inhibitory protein thought to play a role in the stabilization of tuberin. Mutations in this gene have been associated with tuberous sclerosis. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2009]
UniProt Comments for TSC1
TSC1: tuberous sclerosis protein 1 (TSC1). May have tumor suppressor activity. Together with tuberin (TSC2), inhibits mammalian target of rapamycin (mTOR)-mediated signaling to eukaryotic initiation factor 4E-binding protein 1 (4E-BP1) and ribosomal protein S6 kinase. Regulated by the phosphatidylinositol 3-kinase/Akt pathway and phosphorylation of tuberin. Phosphorylated by cyclin-dependent kinase 1/cyclin B during the cell cycle.

1. Data show that microRNA miR-451 target tuberous sclerosis 1 (TSC1) gene and activates the PI3-kinase/Akt protein/mTOR protein signaling in multiple myeloma (MM) Side population (SP) cells.
