NCBI Summary for SOS1
This gene encodes a protein that is a guanine nucleotide exchange factor for RAS proteins, membrane proteins that bind guanine nucleotides and participate in signal transduction pathways. GTP binding activates and GTP hydrolysis inactivates RAS proteins. The product of this gene may regulate RAS proteins by facilitating the exchange of GTP for GDP. Mutations in this gene are associated with gingival fibromatosis 1 and Noonan syndrome type 4. [provided by RefSeq, Jul 2008]
UniProt Comments for SOS1
SOS1: a guanine nucleotide exchange factor that activates Ras by catalyzing the exchange of bound GDP for GTP. Interacts with GRB2, NCK, and other adapter proteins. Plays an important role in signaling pathways leading from the G-protein- coupled receptors and receptor tyrosine kinases to MAPK/ERK.
Protein type: Adaptor/scaffold; GEFs; GEFs, Ras; Motility/polarity/chemotaxis
Chromosomal Location of Human Ortholog: 2p21
Cellular Component: cytosol; plasma membrane
Molecular Function: GTPase activator activity; guanyl-nucleotide exchange factor activity; protein binding; Ras guanyl-nucleotide exchange factor activity; Rho guanyl-nucleotide exchange factor activity
Biological Process: axon guidance; epidermal growth factor receptor signaling pathway; insulin receptor signaling pathway; leukocyte migration; MAPKKK cascade; positive regulation of apoptosis; Ras protein signal transduction; regulation of small GTPase mediated signal transduction
Disease: Fibromatosis, Gingival, 1; Noonan Syndrome 4
Research Articles on SOS1
1. The present study provides a first evidence of allelic imbalance of SOS1 and pinpoints this condition as a possible mechanism underlying a different penetrance of some SOS1-mutated alleles in unrelated carriers
