MBS287540 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the 2-acylglycerol O-acyltransferase 3 (MOGAT3) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MOGAT3. The ELISA analytical biochemical technique of the MBS287540 kit is based on MOGAT3 antibody-MOGAT3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MOGAT3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MOGAT3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Description: This assay employs a two-site sandwich ELISA to quantitate MOGAT3 in samples. An antibody specific for MOGAT3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MOGAT3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MOGAT3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MOGAT3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The deduced 341-amino acid protein contains up to 5 predicted transmembrane domains and has a predicted molecular mass of 36 kD. MOGAT3 shares 49%, 44%, and 46% amino acid sequence identity to human DGAT2, MOGAT1, and MOGAT2, respectively. Fluorescence-tagged MOGAT3 expressed in COS-7 cells localized to the endoplasmic reticulum in a reticular staining pattern indistinguishable from that of calnexin.
MOGAT3 mRNA is expressed almost exclusively in the digestive system, with highest expression in the ileum and lower expression in the remaining portions of the lower gastrointestinal tract and liver. MOGAT3 mRNA was not detected in the stomach, esophagus, trachea, or in any other tissues tested.

Acyl-CoA:monoacylglycerol acyltransferase (MOGAT; EC 2.3.1.22) catalyzes the synthesis of diacylglycerol from 2-monoacylglycerol and fatty acyl-CoA (Cheng et al., 2003 [PubMed 12618427]).[supplied by OMIM, Mar 2008]

Catalyzes the formation of diacylglycerol from 2-monoacylglycerol and fatty acyl-CoA. Also able to catalyze the terminal step in triacylglycerol synthesis by using diacylglycerol and fatty acyl-CoA as substrates. Has a preference toward palmitoyl-CoA and oleoyl-CoA. May be involved in absorption of dietary fat in the small intestine by catalyzing the resynthesis of triacylglycerol in enterocytes.
