This F9 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Guinea pig F9. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team

The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing F9. The ELISA analytical biochemical technique of the MBS733376 kit is based on F9 antibody-F9 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect F9 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, F9. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay: F9 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-F9 antibody and an F9-HRP conjugate. The assay sample and buffer are incubated together with F9-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the F9 concentration since F9 from samples and F9-HRP conjugate compete for the anti-F9 antibody . Since the number of sites is limited, as more sites are occupied by F9 from the sample, fewer sites are left to bind F9-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The F9 concentration in each sample is interpolated from this standard curve.

This gene encodes vitamin K-dependent coagulation factor IX that circulates in the blood as an inactive zymogen. This factor is converted to an active form by factor XIa, which excises the activation peptide and thus generates a heavy chain and a light chain held together by one or more disulfide bonds. The role of this activated factor IX in the blood coagulation cascade is to activate factor X to its active form through interactions with Ca+2 ions, membrane phospholipids, and factor VIII. Alterations of this gene, including point mutations, insertions and deletions, cause factor IX deficiency, which is a recessive X-linked disorder, also called hemophilia B or Christmas disease. F9: Factor IX is a vitamin K-dependent plasma protein that participates in the intrinsic pathway of blood coagulation by converting factor X to its active form in the presence of Ca(2+) ions, phospholipids, and factor VIIIa. Defects in F9 are the cause of recessive X-linked hemophilia B (HEMB); also known as Christmas disease. Mutations in position 43 (Oxford-3, San Dimas) and 46 (Cambridge) prevents cleavage of the propeptide, mutation in position 93 (Alabama) probably fails to bind to cell membranes, mutation in position 191 (Chapel-Hill) or in position 226 (Nagoya OR Hilo) prevent cleavage of the activation peptide.
