The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CCR3. The ELISA analytical biochemical technique of the MBS282669 kit is based on CCR3 antibody-CCR3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CCR3 antigen targets in samples. CCR3 is a receptor for C-C type chemokines. It belongs to family 1 of the G protein-coupled receptors. This receptor binds and responds to a variety of chemokines, including eotaxin (CCL11), eotaxin-3 (CCL26), MCP-3 (CCL7), MCP-4 (CCL13), and RANTES (CCL5). It is highly expressed in eosinophils and basophils, and is also detected in TH1 and TH2 cells, as well as in airway epithelial cells. This receptor may contribute to the accumulation and activation of eosinophils and other inflammatory cells in the allergic airway. It is also known to be an entry co-receptor for HIV-1. This gene and seven other chemokine receptor genes form a chemokine receptor gene cluster on the chromosomal region 3p21. Alternatively spliced transcript variants encoding the same protein have been described.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate CCR3 in samples. An antibody specific for CCR3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCCR3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CCR3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCR3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
