biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing AKAP4. The ELISA analytical biochemical technique of the MBS926560 kit is based on AKAP4 antibody-AKAP4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AKAP4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AKAP4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique.
Antibody specific for AKAP4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AKAP4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AKAP4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AKAP4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The A-kinase anchor proteins (AKAPs) are a group of structurally diverse proteins, which have the common function of binding to the regulatory subunit of protein kinase A (PKA) and confining the holoenzyme to discrete locations within the cell. This gene encodes a member of the AKAP family. The encoded protein is localized to the sperm flagellum and may be involved in the regulation of sperm motility. Alternative splicing of this gene results in two transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]
