ABCB5 (ATP-binding cassette, subfamily B [DR/TAP] member 5) is a 90kD (predicted MW) member of the human P-glycoprotein family of molecules. It is expressed by CD133+ pluripotent pigment stem cells, where it serves as a marker for cells that show a high incidence of polyploidy. Human ABCB5 is 812aa in length. It is possibly a five transmembrane protein with a 247aa N-terminal extracellular domain (ECD) aa1-247 and a 283aa C-terminal cytoplasmic region aa530-812. The ECD contains one ABC transporter aa2-177, while the cytoplasmic region contains a second ABC transporter aa570-808. There are two potential splice forms. One shows a seven aa substitution for aa125-812, while another possesses an alternative start site 445aa upstream of the standard site. Over aa141-247, human ABCB5 shares 64aa identity with mouse ABCB5.

ABCB5 belongs to the ATP-binding cassette (ABC) transporter superfamily of integral membrane proteins. These proteins participate in ATP-dependent transmembrane transport of structurally diverse molecules ranging from small ions, sugars, and peptides to more complex organic molecules (Chen et al., 2005 [PubMed 15760339]).[supplied by OMIM, Mar 2008]

Function: Plasma membrane transporter able to mediate efflux from cells of the rhodamine dye and of the therapeutic drug doxorubicin. Responsible for the resistance to doxorubicin of a subset of malignant melanomas. Ref.1 Ref.4 Ref.10 Ref.11

Subcellular location: Cell membrane; Multi-pass membrane protein Ref.1.

Tissue specificity: Expressed by CD133-expressing progenitor cells among epidermal melanocytes (at protein level). Widely expressed with specific expression in pigment cells. Also expressed in several malignant tissues. Ref.1 Ref.2

Miscellaneous: Depletion of ABCB5 by RNAi increases the sensitivity to several drugs of a subset of melanoma cells.
