Small volumes of cTn-T elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. MBS701710 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Cardiac Troponin T (cTn-T) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing cTn-T. The ELISA analytical biochemical technique of the MBS701710 kit is based on cTn-T antibody-cTn-T antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect cTn-T antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, cTn-T. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to Tn-T has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with biotin-conjugated Tn-T and incubated. A competitive inhibition reaction is launched between Tn-T (Standards or samples) and Biotin-conjugated Tn-T with the pre-coated antibody specific for Tn-T. The more amount of Tn-T in samples, the less antibody bound by Biotin-conjugated Tn-T. Then Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of Tn-T in the sample. The color development is stopped and the intensity of the color is measured.

Tn consists of three components: Tn-I which is the inhibitor of actomyosin ATPase, Tn-T which contains the for tropomyosin and Tn-C. The binding of calcium to Tn-C abolishes the inhibitory action of Tn on actin filaments.
