Specificity
This assay has high sensitivity and excellent specificity for detection of ADORA2a. No significant cross-reactivity or interference between ADORA2a and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ADORA2a and all the analogues, therefore, cross reaction may still exist in some cases.

Principle of the Assay: ADORA2a ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADORA2a antibody and an ADORA2a-HRP conjugate. The assay sample and buffer are incubated together with ADORA2a-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADORA2a concentration since ADORA2a from samples and ADORA2a-HRP conjugate compete for the anti-ADORA2a antibody . Since the number of sites is limited, as more sites are occupied by ADORA2a from the sample, fewer sites are left to bind ADORA2a-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADORA2a concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the guanine nucleotide-binding protein (G protein)-coupled receptor (GPCR) superfamily, which is subdivided into classes and subtypes. The receptors are seven-pass transmembrane proteins that respond to extracellular cues and activate intracellular signal transduction pathways. This protein, an adenosine receptor of A2A subtype, uses adenosine as the preferred endogenous agonist and preferentially interacts with the G(s) and G(olf) family of G proteins to increase intracellular cAMP levels. It plays an important role in many biological functions, such as cardiac rhythm and circulation, cerebral and renal blood flow, immune function, pain regulation, and sleep. It has been implicated in pathophysiological conditions such as inflammatory diseases and neurodegenerative disorders. Alternative splicing results in multiple transcript variants. A read-through transcript composed of the upstream SPECC1L (sperm antigen with calponin homology and coiled-coil domains 1-like) and ADORA2A (adenosine A2a receptor) gene sequence has been identified, but it is thought to be non-coding.
