ALKBH3 recombinant protein
Description: ALKBH3 Human Recombinant fused to 39 amino acid His Tag at N-terminal produced in E Coli is a single, non-glycosylated, polypeptide chain containing 325 amino acids (1-286 a.a.) and having a molecular mass of 37.9kDa. The ALKBH3 is purified by proprietary chromatographic techniques.
Introduction: ALKBH3 belongs to the ALKB family of proteins and acts as a dioxygenase with a preference for RNA and single stranded DNA substrates. ALKBH3 protein repairs 1-methyladenine and 3-methylcytosine lesions in alkylated DNA and RNA and its activity is stimulated by ascorbate. ALKBH3 is expressed in a broad array of tissues and localizes to the cytoplasm and the nucleus.

NCBI Summary for ALKBH3
The Escherichia coli AlkB protein protects against the cytotoxicity of methylating agents by repair of the specific DNA lesions generated in single-stranded DNA. ALKBH2 (MIM 610602) and ALKBH3 are E. coli AlkB homologs that catalyze the removal of 1-methyladenine and 3-methylcytosine (Duncan et al., 2002 [PubMed 12486230]).[supplied by OMIM, Mar 2008]
UniProt Comments for ALKBH3
ALKBH3: Dioxygenase that repairs alkylated DNA containing 1- methyladenine (1meA) and 3-methylcytosine (3meC) by oxidative demethylation. Has a strong preference for single-stranded DNA. Able to process alkylated 3mC within double-stranded regions via its interaction with ASCC3, which promotes DNA unwinding to generate single-stranded substrate needed for ALKHB3. May also act on RNA. Requires molecular oxygen, alpha-ketoglutarate and iron. Belongs to the alkB family. 2 isoforms of the human protein are produced by alternative splicing.
Protein type: Demethylase; DNA repair, damage; Hydrolase; EC 1.14.11.-; Oxidoreductase
Chromosomal Location of Human Ortholog: 11p11.2
Cellular Component: nucleoplasm; mitochondrion; cytoplasm; nucleus
Molecular Function: protein binding; L-ascorbic acid binding; ferrous iron binding; DNA-N1-methyladenine dioxygenase activity; DNA demethylase activity
