The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing IFIH1. The ELISA analytical biochemical technique of the MBS2022859 kit is based on IFIH1 antibody-IFIH1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect IFIH1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, IFIH1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Interferon Induced Helicase C Domain Containing Protein 1 (IFIH1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Interferon Induced Helicase C Domain Containing Protein 1 (IFIH1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Interferon Induced Helicase C Domain Containing Protein 1 (IFIH1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Interferon Induced Helicase C Domain Containing Protein 1 (IFIH1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD), are putative RNA helicases. They are implicated in a number of cellular processes involving alteration of RNA secondary structure such as translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Based on their distribution patterns, some members of this family are believed to be involved in embryogenesis, spermatogenesis, and cellular growth and division. This gene encodes a DEAD box protein that is upregulated in response to treatment with beta-interferon and a protein kinase C-activating compound, mezerein. Irreversible reprogramming of melanomas can be achieved by treatment with both these agents; treatment with either agent alone only achieves reversible differentiation. IFIH1: Innate immune receptor which acts as a cytoplasmic sensor of viral nucleic acids and plays a major role in sensing viral infection and in the activation of a cascade of antiviral responses including the induction of type I interferons and proinflammatory cytokines. Its ligands include mRNA lacking 2'-O- methylation at their 5' cap and long-dsRNA (>1 kb in length).
