Specificity
Mouse anti Human CD16 antibody, clone LNK16 recognizes CD16, a 50-65kDa cell surface molecule which exists in two forms - a transmembranous form expressed by NK cells and some T cells, and a phosphatidylinositol linked form expressed by granulocytes.
CD16 is a low affinity receptor for IgG (FcR III), and is an important receptor mediating ADCC by NK cells. Mouse anti Human CD16 antibody, clone LNK16 has been reported to block the binding of IgG to the CD16 molecule (Tamm and Schmidt 1996).

Storage in frost-free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody.
Should this product contain a precipitate we recommend microcentrifugation before use.
Other Notes
Small volumes of anti-CD16 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Applications Tested/Suitable for anti-CD16 antibody
Flow cytometry (FC/FACS), Immunoprecipitation (IP)
Application Notes for anti-CD16 antibody
FC/FACS - Suggested Dilution: 1/10 - 1/20
Use 10 ul of the suggested working dilution to label 106 cells or 100ul human whole blood.
Where this antibody has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the antibody for use in their own system using appropriate negative/positive controls.

NCBI Summary for CD16
The protein encoded by this gene is a low affinity receptor for the Fc region of gamma immunoglobulins (IgG). The encoded protein acts as a monomer and can bind either monomeric or aggregated IgG. This gene may function to capture immune complexes in the peripheral circulation. Several transcript variants encoding different isoforms have been found for this gene. A highly-similar gene encoding a related protein is also found on chromosome 1. [provided by RefSeq, Aug 2012]
UniProt Comments for CD16
Receptor for the Fc region of immunoglobulins gamma. Low affinity receptor. Binds complexed or aggregated IgG and also monomeric IgG. Contrary to III-A, is not capable to mediate antibody-dependent cytotoxicity and phagocytosis. May serve as a trap for immune complexes in the peripheral circulation which does not activate neutrophils.
Research Articles on CD16
1. In Taiwanese individuals, a low FCGR3A copy number is a common risk factor for systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA), while a low FCGR3B copy number confers a risk of SLE but not RA.
