Description: This assay employs a two-site sandwich ELISA to quantitate HTR7 in samples. An antibody specific for HTR7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HTR7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HTR7 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HTR7 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The receptor is a member of the GPCR superfamily of cell surface receptors and is activated by the neurotransmitter serotonin (5-HT) The 5-HT7 receptor is coupled to Gs (stimulates the production of the intracellular signaling molecule cAMP) and is expressed in a variety of human tissues, particularly in the brain, the gastrointestinal tract, and in various blood vessels. This receptor has been a drug development target for the treatment of several clinical disorders. The 5-HT7 receptor is encoded by the intron containing HTR7 gene, which in humans is transcribed into 3 different splice variants.In humans, the neurotransmitter serotonin (5-HT) plays an important role in various cognitive and behavioral functions. The serotonin receptor encoded by the HTR7 gene belongs to the superfamily of G protein-coupled receptors (GPCRs).
