Product Note
Select online data sheet information is drawn from bioinformatics databases, occasionally resulting in ambiguous or non-relevant product information. It is the responsibility of the customer to review, verify, and evaluate the information to make sure it matches their requirements before purchasing the kit. Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of PTRF elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS282314 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Polymerase I and transcript release factor (PTRF) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PTRF. The ELISA analytical biochemical technique of the MBS282314 kit is based on PTRF antibody-PTRF antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PTRF antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PTRF. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

PTRF elisa kit
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate PTRF in samples. An antibody specific for PTRF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPTRF present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTRF is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTRF bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Summary for PTRF
This gene encodes a protein that enables the dissociation of paused ternary polymerase I transcription complexes from the 3' end of pre-rRNA transcripts. This protein regulates rRNA transcription by promoting the dissociation of transcription complexes and the reinitiation of polymerase I on nascent rRNA transcripts. This protein also localizes to caveolae at the plasma membrane and is thought to play a critical role in the formation of caveolae and the stabilization of caveolins. This protein translocates from caveolae to the cytoplasm after insulin stimulation. Caveolae contain truncated forms of this protein and may be the site of phosphorylation-dependent proteolysis. This protein is also thought to modify lipid metabolism and insulin-regulated gene expression. Mutations in this gene result in a disorder characterized by generalized lipodystrophy and muscular dystrophy. [provided by RefSeq, Nov 2009]
UniProt Comments for PTRF
PTRF: Plays an important role in caveolae formation and organization. Required for the sequestration of mobile caveolin into immobile caveolae. Termination of transcription by RNA polymerase I involves pausing of transcription by TTF1, and the dissociation of the transcription complex, releasing pre-rRNA and RNA polymerase I from the template. PTRF is required for dissociation of the ternary transcription complex. Defects in PTRF are the cause of congenital generalized lipodystrophy type 4 (CGL4). It is a disorder characterized by the association of congenital generalized lipodystrophy with muscular dystrophy and cardiac anomalies. Congenital generalized lipodystrophy is characterized by a near complete absence of adipose tissue, extreme insulin resistance, hypertriglyceridemia, hepatic steatosis and early onset of diabetes. Belongs to the PTRF/SDPR family. 3 isoforms of the human protein are produced by alternative splicing.

Research Articles on PTRF
1. Data suggest that the predominant plasmalemmal anionic lipid phosphatidylserine is essential for proper caveolin/cavin clustering, caveola formation, and caveola dynamics; membrane scrambling (by using other phospholipids) can perturb caveolar stability; these studies involved caveolin-1 and cavin-1 as models of caveolin/cavin clustering.
