◆ CH50 测试（经典通路补体活性）
经典通路法 （CH50） 基于在 Ca++ 和 Mg++ 存在下裂解致敏的绵羊红细胞。
当怀疑补体耗竭或缺乏时，它可以作为筛查试验。 该测试对通路成分的减少、缺失和惰性很敏感。 该方法也适用于评估药品对补体成分的抑制或消耗的影响。
◆ 应用
经典路径补体活性的测定
◆ 原理
将红细胞悬浮液与顺序稀释的血清或血浆在37°C孵育30分钟。
补体系统的活性引起溶血。 孵育后，离心样品得到上清液。 使用波长为415nm的分光光度计测量上清液中游离血红蛋白的浓度，其与补体活性成正比。 绘制血浆稀释因子与溶血程度的关系可以计算CH50。 （即稀释血清以获得 50% 的红细胞裂解）
该试剂盒旨包含少量样品（25 μL）中通过替代途径测量补体活性，并且在 96 孔（圆底）微孔板上是可行的。 也可以使用微量离心管进行测定。
