Description |
The Immortalized Human Osteoprecursor Cells (OPC1) is a conditionally immortalized fetal osetoprecusor cell line uniquely expressing SV40 Tag in the presence of human interferon A/D which affects the proliferation capability. However, the Tag may be expressed constitutively in the presence of serum. It expresses alkaline phosphatase (ALP), osetocalcin (OC), osteonectin (ON), osteopontin (OP), procollagen-I (ProI), and PTH receptor (PTHR). The cells are able to differentiate in the presence of recombinant human BMP-2 (rhBMP2), a regulatory factor involved in differentiation. This cell line is valuable tool as a model system in studies in bone-graft substitutes. |

Propagation |
Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is PriGrow VIII, available at abm, Cat. No. TM018. To make the completed growth medium, add the following components to the base medium: 5% fetal bovine serum (TM999), 750 U/ml human A/D interferon (R&D Systems), 1% Penicillin/Streptomycin (G255), and 0.2 mg/ml G418 for selection. Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.
To differentiate add rhBMP2 at 10 ng/ml. |
