MBS2019524 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Cholinergic Receptor, Muscarinic 3 (CHRM3) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CHRM3. The ELISA analytical biochemical technique of the MBS2019524 kit is based on CHRM3 antibody-CHRM3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CHRM3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CHRM3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cholinergic Receptor, Muscarinic 3 (CHRM3). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Cholinergic Receptor, Muscarinic 3 (CHRM3). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Cholinergic Receptor, Muscarinic 3 (CHRM3), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Cholinergic Receptor, Muscarinic 3 (CHRM3) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The muscarinic cholinergic receptors belong to a larger family of G protein-coupled receptors. The functional diversity of these receptors is defined by the binding of acetylcholine and includes cellular responses such as adenylate cyclase inhibition, phosphoinositide degeneration, and potassium channel mediation. Muscarinic receptors influence many effects of acetylcholine in the central and peripheral nervous system. mAChR M3: The muscarinic acetylcholine receptor mediates various cellular responses, including inhibition of adenylate cyclase, breakdown of phosphoinositides and modulation of potassium channels through the action of G proteins. Primary transducing effect is Pi turnover. Defects in CHRM3 are the cause of Eagle-Barrett syndrome (EGBRS). EGBRS is a syndrome characterized by thin abdominal musculature with overlying lax skin, cryptorchism, megacystis with disorganized detrusor muscle, and urinary tract abnormalities.
